DyNAwax™ Reagent
Improves the specificity and yield of PCRDescription
DyNAwax™ Reagent is designed to improve the specificity and yield of PCR*. The wax separates reaction components in reactions prior to heating. In hot start the reaction components are pipetted in two phases, the wax between them. The bottom phase consists of template, primers, buffer and water. The upper phase above the wax consists of enzyme, nucleotides, buffer and water. Best results are obtained if the phases are approximately equal in size. The DyNAwax Reagent can also be used to replace mineral oil in the PCR* to prevent evaporation of reaction mixtures at high temperatures. In such cases the use of wax is similar to that of mineral oil on top of the reaction mixture. The wax melts at 60 – 70 °C (e.g. in a water bath). Two to three drops of hot wax are pipetted using a hot pipette tip onto the bottom phase. When the wax has solidified the upper phase can be pipetted on top of the wax. The wax solidifies on ice almost immediately. After the PCR* reaction it is advisable to pierce the wax with a separate tip before pipetting the reaction mixture out of the reaction tube.
